Mechanism of action

Senescent Cell Specificity: In senescent cells, the FOXO4 protein aberrantly binds and sequesters p53 in the nucleus, inhibiting its normal pro-apoptotic function.

Competitive Binding: The FOXO4-DRI peptide competitively binds to FOXO4, releasing p53.

Restoration of Apoptosis: The released p53 can then activate downstream pro-apoptotic target genes (e.g., PUMA, BAX), initiating the selective apoptosis of senescent cells.

Effect on Normal Cells: In normal cells, where p53 is primarily cytoplasmic and FOXO4-p53 interaction is minimal, the peptide has negligible effects.

In summary, this peptide acts as a "molecular key" that specifically unlocks the inhibited p53 in senescent cells, restarting their apoptotic program.

Usage Guidelines (Research UseOnly):
